Would this be for an image host type of setup? What kind of files are being uploaded?

You would most likely need a MySQL/PHP combo setup. Could be on a linux or windows server, what ever you're more comfortable with.

Why would they be as large as 2gb if they are not multimedia?

it would be for image files.
but would mysql/php be able to handle well such traffic and demand?
would you recommend having the mysql on a second box or the same as the web one?

also would anyone have a recommendation for a templated engine that would already be prebuild for php-mysql transactions...
I'm looking to setup someting that would not require me to do too much developement as I'm not that good at it!

MySQL/PHP will be fine. Your server setup will depend on the amount of traffic you expect. You'd most likely want to go with either a managed VPS or managed dedicated server setup depending on the amount of resources you presume will be utilized.
