I have quite a few desktops (XP) to look after. Most of these got a basic load one time and that is it. No program uninstalls, reinstalls. Just "day to day" use. Is it worth the time spent to run a defrag on these?

I'd say yes if you have the time to do it. Even on XP and Vista, it's a useful tool to clean up the drive and restore some efficiency to it. I wouldn't do it but every month, but not any more than that.

Let the defrag application analyse the disk & follow its' reccomendation.
On a day-to-day system that has the same usage patterns every day I usually get by about 3 months before the system reccomends following up with the actual defrag.

I had a 120gb harddisk for my weekly backup. defrag it once every 2-3 months to bring up back to speed and cut down the defragmentation.

I'd say yes, I defrag every week to keep in clean and healthy. And what is exactly 'day-to-day' use?

Is it posible to make a policy or write a script to force all computers in the domain to run a defrag?

Here's what I did....... the round-a-bout way.

1. Created a scheduled task on my own computer exactly how I want it to run on all the others. (this must be run as an administer)
2. Copied that task to a folder on a network drive.
3. Wrote a short .vbs to copy that scheduled task to the Scheduled Task folder on the local computer.
4. Created a Startup Group Policy to run the script after a login. (this is when the file is copied)
5. I rebooted my compter, and after logging in I checked the scheduled tasks and the Defrag one was now there.
