Right click on the drive in question.
Select 'Properties' <-- I forget this step
Go to the security tab
Click 'Add'
Enter this in the text field: Administrators
Click 'OK'
Under 'Allow', check 'Full Control'

Click OK

That should do it.

Go back to the security tab for that drive, select Everyone, Under Allow, check 'Read & execute'. Click Ok.

Try this:

On the security tab, click 'Advanced'
Go to the Owner tab
Select 'Administrators'
Check 'Replace Owner on Subcontainers and objects'
Click 'OK'
When prompted, click yes.

Hopefully that will enable access.

Glad you got it sorted. If you can access your files then I would say it's working. No reason why it would stop working under normal circumstances. I can't possibily predict what may go wrong in the future.

*smiles -- yep, I did warn her:

That was the first thing I learned. You simply cannot set everyone to "deny" and expect to access the drive. I'm also glad to see you got it sorted. I got stuck on a plane for the last 9 hours and couldn't get back to help. Learning how to set permissions properly is a pain in the arse, but once you've done it a few times it gets pretty easy.

The inheritance is the hardest part to understand, but as long as you keep the key rule in mind that deny overrides allow you won't go wrong. This is also important to remember if you use groups. Even though you may be an admin, if your admin is part of a group and you deny that group, you will lose rights to the access even though as an individual user you have full allow.

Permissions for Windows is retarded! It should be a top-down design where permissions on top override permissions listed below or vice-versa. Ok....my venting is done now.
